Expression of GABA and its receptors in the rat spinal dorsal horn following peripheral inflammatory hyperalgesia MENG Chun*,WANG Guo-lin. Department of Anesthesiology, Tianjin Medical University General Hospital, Tianjin 300052, *Tianjin Huanhu Hospital, Tianjin 300060, China
【Abstract】Objective To study the changes of the expression of GABA and its receptors in spinal dorsal horn of rats in formalin test. Methods The rats were randomly divided into two groups: six non-stimulation rats were used as control(A); six rats were subcutaneously injected 5% formalin 50μl into the right hindpaw to produce persistent nociception(B). Rats were killed after 24h respectively. Using the immunocytochemistry method, the changes of the number of GABA immunoreactive cells were observed, and the expression of the GABAAβ3 and GABAB1 receptor mRNA in the spinal dorsal horn were determinded by in situ hybridization. Results GABA immunoreactive cells and GABAB1 receptor mRNA were observed throughout the lumbar spinal dorsal horn over cell bodies with the highest density in superficial laminae Ⅰ~Ⅲ of the normal rats, and GABAAβ3 receptor mRNA was well-distributed in spinal dorsal horn compared with GABA immunoreactive cells and GABAB1 receptor mRNA. Twenty-four hours after formalin injection, a significant increase of the GABA-immunoreactive cells in ipsilaterally spinal dorsal horn was observed, the expression of GABAAβ3 and GABAB1 receptor mRNA were increased in bilateral spinal dorsal horn compared with that of A group, three parameters were positive correlation and their location of the expression in spinal dorsal horn didn’t change compared with that of A group.
Conclusion GABA and its receptors may play an important role in regulating nociceptive modulation.

结果观察和阳性指标计数 在高倍光学显微镜下,选取脊髓背角不重叠的4个高倍视野(400倍),计数视野范围内的阳性细胞数,计算其平均值。免疫组化阳性细胞胞浆呈棕黄色,胞核不着色(复染后呈蓝色),背景淡黄色,阴性细胞胞浆不着色。原位杂交阳性细胞胞浆呈棕黄色,胞核不着色(复染后呈蓝色),背景淡黄色,阴性细胞胞浆不着色。
统计学处理 计量资料以均数±标准差( )表示,以SPSS11.0进行统计分析。同鼠致痛侧与非致痛侧比较应用配对t检验,组间比较应用单因素方差分析、q检验和相关分析。以P<0.05为差异有显著性。
结 果
定位结果 实验各组脊髓腰段背角GABA免疫阳性细胞、GABAAβ3和GABAB1受体mRNA阳性细胞胞浆呈棕黄色,复染后核呈蓝色,背景淡黄色。阳性细胞分布于脊髓各层,密度存在差异:GABA免疫阳性细胞在脊髓背角第Ⅰ~Ⅱ层分布最为密集,在第Ⅳ~Ⅵ层、Ⅹ层和前角分布稀疏(图1~2);GABAAβ3受体mRNA阳性细胞在脊髓各层分布较均匀(图3~4);GABAB1受体mRNA阳性细胞在脊髓背角Ⅰ~Ⅲ层密度最高,第Ⅳ~Ⅷ层和第Ⅹ层内也可见其分布(图5~6)。无论是在A组还是B组,上述三个指标的形态变化不大,在脊髓层面的分布相似,但是阳性细胞数量存在差异。
