The Value of NR2B Subunit of NMDA Receptor in Pain Therapy<?xml:namespace prefix = o ns = "urn:schemas-microsoft-com:office:office" /> 林培容 综述 ABSTRACT The NR2B-containing NMDA receptors are implicated in pain perception. NR2B antagonists are antinociceptive in a variety of preclinical pain models and have a much lower side-effect profile compared with other NMDA receptor antagonists. Human trials have so far confirmed the good tolerability of these subtype-selective NMDA receptor antagonists; However, whether they are as effective as other NMDA receptor antagonists in pain patients remains to be demonstrated. 谷氨酸是中枢神经系统最重要的兴奋性神经递质,通过一系列离子型或代谢型膜受体发挥其突触后作用。NMDA受体是谷氨酸受体的一种,与中枢的许多功能如认知、记忆、神经元可塑性以及疼痛信号转导均有关,含有NR2B的NMDA受体对伤害性信号传递尤其重要。NR2B对神经元兴奋性具有正性的调节作用,与其他的镇痛药物相比,选择性作用于NR2B的NMDA受体拮抗剂镇痛效果显著,同时不会导致耐受、精神性副反应,将是未来镇痛药物发展的方向之一。 |
